目的研究乳腺导管癌中MMP-2、MMP-9与ColⅣ的表达及其相关性,并探讨其与临床病理参数之间的关系。方法应用免疫组化双染法检测60例乳腺导管癌中MMP-2、MMP-9和ColⅣ的表达,并进行相关性分析,同时分析MMP-2、MMP-9与乳腺导管癌患者的年龄、肿瘤大小、淋巴结转移及肿瘤ER、PR、E-cad、C-erbB-2的关系。结果 MMP-2、MMP-9在乳腺导管癌中的表达显著高于乳腺正常组织(P<0.05),而ColⅣ在乳腺癌组织中的表达低于乳腺正常组织(P<0.05)。MMP-2、MMP-9在靠近基膜或者于将要突破基膜处的癌细胞中表达较强。MMP-2、MMP-9在不同年龄组、PR、ER、E-cad分组间的表达无显著差异(P>0.05);但在肿瘤直径>2 cm、有淋巴结转移及C-erbB-2阳性组的表达显著高于相应的对照组(P<0.05)。ColⅣ的表达与MMP-2的表达呈负相关(r=-0.506,P=0.000),亦与MMP-9的表达呈负相关(r=-0.501,P=0.000)。结论 MMP-2、MMP-9对ColⅣ的降解可能在乳腺导管癌的局部侵袭和远处转移过程中发挥重要作用。MMP-2、MMP-9和ColⅣ的表达,可能对判断乳腺癌的恶性程度和浸润转移潜能以及评估预后有参考价值。
Objective To study the expression and correlation of MMP-2, MMP-9 and ColⅣ in breast ductal carcinoma and to explore its relationship with clinicopathological parameters. Methods The expressions of MMP-2, MMP-9 and ColⅣ in 60 cases of ductal carcinoma of the breast were detected by immunohistochemistry and double staining. The correlation analysis was made between the expressions of MMP-2, MMP-9 and the age, Tumor size, lymph node metastasis and tumor ER, PR, E-cad, C-erbB-2 relationship. Results The expression of MMP-2 and MMP-9 in breast ductal carcinoma was significantly higher than that in normal breast tissue (P <0.05), while the expression of ColⅣ in breast cancer was lower than that in normal breast tissue (P <0.05). MMP-2 and MMP-9 are highly expressed in cancer cells near the basement membrane or at the basement membrane. There was no significant difference in the expression of MMP-2 and MMP-9 between PR, ER and E-cad groups in different age groups (P> 0.05). However, in the group of tumor diameter> 2 cm, lymph node metastasis and C-erbB- Group was significantly higher than the corresponding control group (P <0.05). The expression of ColⅣ was negatively correlated with the expression of MMP-2 (r = -0.506, P = 0.000) and negatively correlated with the expression of MMP-9 (r = -0.501, P = 0.000). Conclusion The degradation of ColⅣ by MMP-2 and MMP-9 may play an important role in the local invasion and distant metastasis of breast ductal carcinoma. The expression of MMP-2, MMP-9 and ColⅣ may be valuable in judging the malignancy, invasion and metastasis potential of breast cancer and prognosis.
