目的观察肾上腺髓质素(AM)对高糖培养的肾小球系膜细胞HBZY-1增殖和凋亡的影响及其可能的作用机制。方法采用MTT法和流式细胞仪测定观察AM干预对HBZY-1细胞增殖和凋亡的影响,同时采用Western blot观察分裂素激活蛋白激酶(MAPK)信号传导途径中ERK和P38MAPK蛋白活性的变化。结果高糖状态下HBZY-1细胞增殖明显增强,凋亡轻度增加;AM干预后HBZY-1细胞增殖明显抑制,平均下降38.8%,同时凋亡明显增强。AM干预后,ERK1/2活性明显抑制,与高糖组比较平均下降约30%,而P38MAPK活性明显增强,较高糖组平均增高约70%。结论AM对高糖培养HBZY-1细胞增殖增强具有抑制作用,并促进其凋亡。这种作用是可能是通过MAPK信号传导途径所介导的。
Objective To observe the effects of adrenomedullin (AM) on the proliferation and apoptosis of cultured mesangial cells (HBZY-1) induced by high glucose and its possible mechanism. Methods MTT and flow cytometry were used to observe the effect of AM on proliferation and apoptosis of HBZY-1 cells. Western blot was used to observe the changes of ERK and P38MAPK proteins in mitogen-activated protein kinase (MAPK) signal transduction pathway. Results The proliferation of HBZY-1 cells was significantly increased and the apoptosis increased slightly under high glucose conditions. The proliferation of HBZY-1 cells was inhibited by AM intervention, with an average decrease of 38.8% and the apoptosis of HBZY-1 cells significantly increased. After AM intervention, ERK1 / 2 activity was significantly inhibited, with an average reduction of about 30% compared with high glucose group, while P38MAPK activity was significantly increased, an average increase of about 70% higher glucose group. Conclusion AM can inhibit the proliferation of HBZY-1 cells cultured in high glucose and promote its apoptosis. This effect is probably mediated through the MAPK signaling pathway.
