目的构建转移抑制基因nm23-H_1cDNA克隆,初步分析其mRNA表达与肝内转移、门静脉形成癌栓、TNM分期等临床病理特征的关系。方法利用RT-PCR技术扩增nm23-H_1cDNA片段,并亚克隆人pBluescript Ⅱ SK(-)质粒,借助Northern blot方法,检测10例肝细胞癌及癌旁肝组织nm23-H_1基因mRNA表达水平。结果 nm23-H_1mRNA低表达,诱发肝细胞癌伴肝内转移和/或门静脉癌栓形成的机会增加,nm23-H_1 可能也是肝癌转移抑制基因。结论 nm23-H_1基因mRNA低表达,在肝癌浸润转移过程中起着重要的作用。
Objective To construct the nm23-H_1 cDNA clone of metastasis suppressor gene and analyze its relationship with the clinicopathological features such as intrahepatic metastasis, portal vein thrombosis, and TNM staging. Methods RT-PCR was used to amplify the nm23-H1 cDNA fragment, and the human pBluescript II SK(-) plasmid was subcloned. Northern blot was used to detect the mRNA expression of nm23-H1 mRNA in 10 cases of hepatocellular carcinoma and adjacent liver tissues. Results The low expression of nm23-H_1 mRNA increased the chance of hepatocellular carcinoma with intrahepatic metastasis and/or portal vein tumor thrombus formation. nm23-H_1 may also be a metastasis suppressor gene of hepatocellular carcinoma. Conclusion The low expression of nm23-H1 mRNA plays an important role in the invasion and metastasis of HCC.
