目的:建立艾滋病口腔含漱液中有效成分苦参碱与氧化苦参碱的含量测定方法。方法:采用高效液相色谱法,色谱柱Agilent ZORBAX NH2(250 mm×4.6 mm,5μm),流动相为乙腈-无水乙醇-3%磷酸溶液(82∶9∶9),流速:1.0 ml·min-1,检测波长:222 nm,柱温:30℃,进样量:10μl。结果:苦参碱在6.6~41.3μg·ml-1(r=0.999 6)、氧化苦参碱在8.2~51.2μg·ml-1(r=0.999 6)的浓度范围内线性关系良好,平均回收率分别为99.96%、99.70%,RSD分别为1.46%,1.29%(n=6)。结论:本方法操作简便、重复性好,可以更有效地控制制剂的质量。
Objective: To establish a method for the determination of the active ingredients matrine and oxymatrine in the oral cavity gargle of AIDS. Methods: High performance liquid chromatography with Agilent ZORBAX NH2 column (250 mm × 4.6 mm, 5 μm) was used. The mobile phase was acetonitrile-absolute ethanol-3% phosphoric acid solution min-1, detection wavelength: 222 nm, column temperature: 30 ℃, injection volume: 10μl. Results: The linear range of matrine was 6.6 ~ 41.3μg · ml-1 (r = 0.999 6) and oxymatrine was 8.2 ~ 51.2μg · ml-1 (r = 0.999 6) Rates were 99.96% and 99.70%, respectively, with RSDs of 1.46% and 1.29%, respectively (n = 6). Conclusion: The method is simple, reproducible and can control the quality of the preparation more effectively.
