目的 :探讨携带人肝细胞生长因子基因的质粒pUDKH对 13日龄鸡胚绒膜尿囊膜血管生成的刺激效应。方法 :以甲基纤维素为载体 ,将不同剂量的pUDKH(1.0 ,2 .0 ,5 .0 μg)或空白质粒pUDK(5 .0 μg)置入 13日龄鸡胚绒膜尿囊膜上 ,于孵化箱继续孵育 3d后 ,观察鸡胚绒膜尿囊膜上小血管的密度。结果 :应用质粒pUDKH各组小血管的密度均明显较对照组大 (P <0 .0 5 ) ,且有剂量_效应关系。结论 :质粒pUDKH可显著刺激鸡胚绒膜尿囊膜上小血管的形成 ,故认为其具有在体内应用于治疗缺血性疾病的潜能
Objective: To investigate the stimulatory effect of plasmid pUDKH carrying human hepatocyte growth factor gene on the angiogenesis of chick chorioallantoic membrane on 13-day-old chickens. Methods: Different doses of pUDKH (1.0, 2.0, 5.0 μg) or blank plasmid pUDK (5.0 μg) were placed on chick embryo chorioallantoic membrane with methyl cellulose as carrier. After incubating for 3 days, the density of small blood vessels in chick embryo chorioallantoic membrane was observed. Results: The density of small vessels in each group of plasmid pUDKH was significantly higher than that of control group (P <0.05), and there was dose-effect relationship. CONCLUSION: Plasmid pUDKH can significantly stimulate the formation of small blood vessels in the chick chorioallantoic membrane and is therefore considered to have the potential to be used for the treatment of ischemic diseases in vivo
