目的 探讨forskolin和IBMX促进受损视网膜节细胞 (RGCs)再生的作用机制。 方法 采用荧光逆行示踪标记术和自体坐骨神经移植术 ,研究蛋白激酶A(PKA)抑制剂H 89或 /和PI3 K特异抑制剂wortmannin玻璃体内注射对forskolin和IBMX促进成年金黄地鼠受损RGCs再生的影响。结果 H 89和wortmannin均可部分抑制forskolin和IBMX对受损RGCs的促再生作用 ,wortmannin的抑制作用更大 ;两药联合应用 ,可完全抑制forskolin和IB MX对受损RGCs的促再生作用。 结论 forskolin和IBMX可能是通过PKA CREB和PI3 K Akt通路实现对受损RGCs的促再生作用
Objective To investigate the mechanism of forskolin and IBMX in promoting the regeneration of injured retinal ganglion cells (RGCs). Methods Fluorescent retrograde tracing and autologous sciatic nerve grafting were used to investigate the effect of forskolin and IBMX on RGCs regeneration in adult golden hamster after intravitreous injections of protein kinase A (PKA) inhibitor H89 and / or PI3K-specific inhibitor wortmannin Impact. Results Both H89 and wortmannin could partially inhibit the pro-regenerative effect of forskolin and IBMX on injured RGCs, and the inhibitory effect of wortmannin was greater. The combined application of both drugs could completely inhibit the pro-regenerative effect of forskolin and IB MX on damaged RGCs. Conclusions Forskolin and IBMX may promote regeneration of impaired RGCs via PKA CREB and PI3K Akt pathways
