目的:探讨同型半胱氨酸(homocysteine,Hcy)对促进血管平滑肌细胞(vascularsmoothmusclecells,VSMCs)增殖的影响。方法:常规培养人脐静脉VSMCs,将培养的VSMCs分为0、50、100、200、500及1000μmol/LHcy,其中0μmol/LHcy组为对照组。当VSM-Cs细胞长满80%瓶底时,各组加入不同浓度的Hcy,继续培养细胞6h、12h、24h、36h、48h、72h后,采用倒置显微镜观察细胞的生长状态,采用MTT法检测细胞的增殖活力。结果:当Hcy刺激VSMCs12~48h时,VSMCs的细胞数逐渐增加,细胞的活力也持续增加(P<0.05);当Hcy的刺激时间达72h时,高浓度(1000μmol/LHcy)组的VSMCs活力的升高趋势有所减缓,细胞有损害表现。结论:高浓度Hcy可直接诱导VSMCs增殖;但随着刺激时间的增加,过高浓度的Hcy对VSMCs表现出抑制和损害效应。
Objective: To investigate the effect of homocysteine (Hcy) on the proliferation of vascularsmooth cells (VSMCs). Methods: Human umbilical vein endothelial cells (VSMCs) were cultured routinely. The cultured VSMCs were divided into 0, 50, 100, 200, 500 and 1000μmol / LHcy groups, 0μmol / LHcy group as control group. When VSM-Cs cells covered 80% of the bottom of the bottle, Hcy was added into each group at different concentrations for 6 h, 12 h, 24 h, 36 h, 48 h, 72 h later. The growth of cells was observed by inverted microscope. Cell proliferation activity. RESULTS: When Hcy stimulated VSMCs for 12 ~ 48h, the number of VSMCs increased and the viability of VSMCs increased (P <0.05). When Hcy stimulated for 72h, VSMCs in high concentration (1000μmol / LHcy) Increasing trend has slowed down, cell damage performance. CONCLUSION: High concentrations of Hcy can directly induce the proliferation of VSMCs. However, with increasing stimulation time, high concentrations of Hcy can inhibit and impair VSMCs.
