用PCR-SSCP分析及DNA直接测序法检测了16例胰腺腺癌的P53基因5~8外显子,结果发现37.5%(6/16)有P53基因点突变,突变分布于外显子5~8,无特殊突变倾向点。6例有P53突变者,5例为1个等位基因突变,另1个等位基因丢失;1例则同时含有突变型和野生型等位基因。DNA测序显示,突变属于碱基转换者5例,其中G到A4例,A到G1例;属于碱基颠换者1例,为A到C替代;突变点位CpG二核工苷酸位者1例。结果提示胰腺腺癌中,P53基因的突变是一个较常见的现象,抗癌基因P53的失活可能在胰腺腺癌的发生发展中起着一定作用。
The P53 gene exons 5~8 in 16 cases of pancreatic adenocarcinoma were detected by PCR-SSCP analysis and DNA direct sequencing. 37.5% (6/16) of the P53 gene point mutations were found, and mutations were found in exons. 5-8, no special tendency to mutation. Six cases had P53 mutations, 5 cases had one allele mutation and the other allele was lost. One case contained both mutant and wild-type alleles. DNA sequencing showed that the mutation belonged to the base conversion in 5 cases, including G to A4 cases, A to G1 cases; belong to the base transposition in 1 case, which was A to C substitution; the mutation site was CpG dinucleotide nucleotides 1 case. The results suggest that mutation of P53 gene is a common phenomenon in pancreatic adenocarcinoma. The inactivation of anticancer gene P53 may play a role in the development of pancreatic adenocarcinoma.
