目的探讨白细胞介素18(IL-18)、肿瘤坏死因子α(TNF-α)对3T3-L1脂肪细胞、RAW264.7巨噬细胞的白细胞介素18受体β(IL-18Rβ)表达的影响。方法采用反转录PCR和Western blot法检测不同浓度IL-18、TNF-α处理后3T3-L1脂肪细胞、RAW264.7巨噬细胞的IL-18RβmRNA及蛋白的表达。结果 10 ng/m L、100 ng/m L TNF-α显著增加3T3-L1脂肪细胞IL-18Rβ的mRNA及蛋白表达,10 ng/m L、100 ng/m L IL-18显著增加RAW264.7巨噬细胞IL-18Rβ的mRNA及蛋白表达。结论IL-18、TNF-α可能通过促进巨噬细胞、脂肪细胞IL-18R的表达参与糖尿病和肥胖的脂肪组织慢性炎症过程。
Objective To investigate the effects of interleukin 18 (IL-18) and tumor necrosis factor α (TNF-α) on the expression of interleukin 18 receptorβ (IL-18Rβ) in 3T3-L1 adipocytes and RAW264.7 macrophages . Methods The mRNA and protein expressions of IL-18Rβ in 3T3-L1 adipocytes and RAW264.7 macrophages treated with different concentrations of IL-18 and TNF-α were detected by reverse transcription-polymerase chain reaction and Western blot. Results The mRNA and protein expressions of IL-18Rβ in 3T3-L1 adipocytes were significantly increased by 10 ng / mL and 100 ng / mL of TNF-α, while 10 ng / mL and 100 ng / mL of IL-18 significantly increased RAW264.7 The mRNA and protein expression of IL-18Rβ in macrophages. Conclusion IL-18 and TNF-α may be involved in the chronic inflammatory process of adipose tissue and adipose tissue by promoting the expression of IL-18R in macrophages and adipocytes.
