目的观察曲安奈德(triamcinoloneacetonide,TA)对培养的人视网膜色素上皮细胞(ARPE19)增生的影响。设计实验性研究。研究对象ARPE19。方法采用MTT比色法和TUNEL染色法观察不同终质量浓度(0、10、30、300mg/L)TA分别作用于培养的ARPE19细胞24、72小时后细胞的增生和凋亡的情况。主要指标吸光度,凋亡指数。结果(1)TUNEL法质量浓度10mg/LTA不影响ARPE19细胞的生长,随着质量浓度增大,细胞凋亡程度逐渐增强(t=2.921,P<0.01),随着时间延长,ARPE19细胞的凋亡指数逐渐增大(t=2.306,P<0.01)。(2)MTT法TA浓度和作用时间均对ARPE19细胞的增生具有显著影响,随着浓度增加和时间延长,其抑制作用的差别有显著性意义(F0.05,3,16=3.24,P<0.05;F0.05,1,16=4.49,P<0.05),而且,TA浓度和作用时间相互影响着ARPE19细胞的生长,对其抑制作用具有统计学意义(F0.05,1,16=4.49,P<0.05)。结论TA能够抑制ARPE19细胞的增生,通过玻璃体腔内注射TA或联合玻璃体切割手术可能成为增生性玻璃体视网膜病变的有效治疗。
Objective To observe the effect of triamcinolone acetonide (TA) on the proliferation of cultured human retinal pigment epithelial cells (ARPE19). Design experimental research. Study object ARPE19. Methods MTT assay and TUNEL staining were used to observe the proliferation and apoptosis of ARPE19 cells cultured in different concentrations (0, 10, 30, 300 mg / L) for 24 and 72 hours respectively. The main indicators of absorbance, apoptosis index. Results (1) The TUNEL concentration of 10 mg / LTA did not affect the growth of ARPE19 cells. With the increase of mass concentration, the apoptosis of ARPE19 cells gradually increased (t = 2.921, P <0.01) The index of death increased gradually (t = 2.306, P <0.01). (2) MTT concentration and duration of action of TA have a significant effect on the proliferation of ARPE19 cells, with the increase of concentration and time, the difference was significant (F0.05,3,16 = 3.24, P < 0.05, F0.05, 1, 16 = 4.49, P <0.05). Moreover, the concentration of TA and the interaction time affected the growth of ARPE19 cells with statistical significance (F0.05,1,16 = 4.49 , P <0.05). Conclusions TA can inhibit the proliferation of ARPE19 cells and may be an effective treatment for proliferative vitreoretinopathy through intravitreal injection of TA or combined vitrectomy.
