目的观察不同剂量的β淀粉样蛋白对大鼠肾上腺嗜铬瘤(PC12)细胞损伤,构建阿尔茨海默病(AD)体外模型。方法将PC12细胞分为正常组,10μmol/L,50μmol/L及100μmol/L三种不同浓度β淀粉样蛋白组,通过形态学和四甲基偶氮唑盐比色法为指标,观察细胞损伤的作用。结果三种不同浓度β淀粉样蛋白组,PC12细胞均有明显损伤。结论β淀粉样蛋白对PC12细胞的损伤随着剂量增加而加重,50μmol/L可作为理想的阿尔茨海默病体外模型。
Objective To observe the injury of rat adrenal pheochromocytoma (PC12) cells with different doses of β-amyloid protein and establish an in vitro model of Alzheimer’s disease (AD). Methods PC12 cells were divided into normal group, 10μmol / L, 50μmol / L and 100μmol / L amyloid β-amyloid groups. Morphological and MTT assay were used to observe cell injury Role. Results Three different concentration of amyloid β group, PC12 cells were significantly damaged. Conclusion The damage of β-amyloid to PC12 cells aggravates with the increase of dose, and 50 μmol / L can be used as an ideal in vitro model of Alzheimer’s disease.
