目的:优化薏苡仁多糖提取工艺。方法:超声波辅助酸水提取,采用蒽酮硫酸法测定多糖含量,以多糖提取率和干浸膏得率为指标,以提取温度、p H值、料液比、时间和次数为考察因素。采用正交试验优选提取工艺,并采用冷冻干燥制备干浸膏。通过极差分析、方差分析,分析各因素对综合评价的影响,确定最佳工艺。结果:优选工艺为10倍量p H 5.0盐酸溶液浸泡10 min,80℃超声波15 min,提取2次。该条件下多糖提取率为14.12%,干浸膏得率为14.45%。结论:优化后的提取工艺经济高效、稳定可行,同时解决了因淀粉高温糊化带来的提取和含量测定环节的干扰。
Objective: To optimize the polysaccharide extraction process of Coix Coats. METHODS: Ultrasonic assisted acid extraction was used to determine the polysaccharide content using anthrone-sulfuric acid method. The polysaccharide extraction rate and dry extract yield were taken as indexes, and the extraction temperature, p H value, material-to-liquid ratio, time and number of times were taken as the investigation factors. Orthogonal experiments were used to optimize the extraction process and dry extracts were prepared using freeze-drying. Through range analysis and variance analysis, the influence of various factors on comprehensive evaluation was analyzed to determine the best process. RESULTS: The optimal technique was to soak the sample in 10-fold pH 5.0 hydrochloric acid solution for 10 min, sonicate at 80 °C for 15 min, and extract twice. The polysaccharide extraction rate was 14.12% under this condition, and the dry extract yield was 14.45%. Conclusion: The optimized extraction process is cost-effective, stable, and feasible, and at the same time solves the interference caused by starch extraction and content determination due to high-temperature gelatinization.
