目的研究温阳活血解毒复方(WHJF)对人脐静脉内皮细胞(HUVECs)增殖、迁移及细胞管形成功能的影响,从细胞层面探讨该治疗动脉粥样硬化机制。方法将SD大鼠随机分为空白对照组、温阳活血解毒复方低、中、高剂量组(剂量分别为10.35,31.05,93.15 g·kg~(-1)),于第4天首次灌胃2 h后采血制备含药血清,采用CCK8法、Transwell迁移实验、b FGF诱导细胞管形成实验分别观察含药血清对HUVECs增殖、迁移及细胞管形成功能的影响。结果温阳活血解毒复方含药血清对HUVECs的增殖有抑制作用,与空白对照组比较,低、中、高剂量组的含药血清对HUVECs的抑制率均明显升高(P<0.05),且两两比较差异有统计学意义(P<0.05),呈浓度依赖型。在迁移方面,温阳活血解毒复方组迁移细胞数减少,与空白对照组比较,高剂量组差异有统计学意义(P<0.05)。在细胞管形成方面,与模型组比较,温阳活血解毒复方组细胞管数明显减少,差异均有统计学意义(P<0.05);与空白对照组比较,低、高剂量组差异有统计学意义(P<0.05)。结论温阳活血解毒复方含药血清可抑制HUVECs的增殖、迁移及细胞管形成,起到稳定动脉粥样硬化斑块的作用;此外,其可能与b FGF产生协同作用,共同促进新生血管成熟,发挥稳定斑块的作用。
Objective To investigate the effect of WHJF on the proliferation, migration and cell tube formation of human umbilical vein endothelial cells (HUVECs), and to explore the mechanism of atherosclerosis from the cellular level. Methods SD rats were randomly divided into blank control group, Wenyang Huoxue Jiedu compound low, medium and high dose groups (doses were 10.35,31.05,93.15 g · kg -1) Blood samples were collected 2 h later to prepare drug-containing serum. The effects of drug-containing serum on the proliferation, migration and cell tube formation of HUVECs were observed by CCK8 assay, Transwell migration assay and b FGF-induced cell tube formation assay. Results Compared with the blank control group, the serum containing Hanyang Huoxue Jiedu Compound could inhibit the proliferation of HUVECs. Compared with the blank control group, the inhibitory rates of HUVECs in the low, medium and high dose groups were significantly increased (P <0.05) The difference between each other was statistically significant (P <0.05), was concentration-dependent. In terms of migration, the number of migrating cells in Wenyang Huoxue Huoxue Jiedu compound group decreased, compared with the blank control group, the difference was statistically significant (P <0.05). In the aspect of cell tube formation, compared with the model group, the number of cells in the group of Wenyang Huoxue Jiedu decoction decreased significantly (P <0.05), compared with the blank control group, there were statistically significant differences between the low and high dose groups Significance (P <0.05). Conclusion Wenyang Huoxue Jiedu compound-containing serum can inhibit the proliferation, migration and cell tube formation of HUVECs and play the role of stabilizing atherosclerotic plaque. In addition, it may have a synergistic effect with b FGF to promote neovascularization, Play a stable plaque role.
