目的评价脱氧胆酰酪氨酸热敏凝胶剂(DCT gel)的小型猪阴道黏膜刺激性。方法将9只雌性小型猪随机分为空白胶组、DCT-gel组和市售壬苯醇醚凝胶(N-9 jelly)组,阴道给予5 ml凝胶,每日1次,连续给药7 d,末次给药72 h后处死小型猪,取阴道组织,观察阴道黏膜组织病理学改变;免疫组化法检测阴道黏膜内CD45的表达。结果小型猪阴道黏膜组织HE染色显示DCT gel对黏膜上皮层、固有层、血管及周围组织无明显损伤(5.5±2.7),与空白胶组相比差异无统计学意义(3.3±1.6),远低于N-9 jelly组(20.5±3.5,P<0.05);与空白胶组相比,DCT gel组的阴道黏膜CD45表达差异无统计学意义(P>0.05),N-9 jelly组CD45表达显著增加(P<0.05)。结论 DCT gel对小型猪阴道黏膜刺激性较小,作为阴道外用杀微生物剂具有一定的应用前景。
Objective To evaluate the vaginal mucosal irritation of minipigs in DCT gel. Methods Nine female miniature pigs were randomly divided into blank gel group, DCT-gel group and N-9 jelly group. 5 ml gel was vaginally administered once daily for continuous administration At 7 days, the piglets were sacrificed 72 hours after the last administration, vaginal tissues were taken and pathological changes of vaginal mucosa were observed. The expression of CD45 in the vaginal mucosa was detected by immunohistochemistry. Results HE staining of vaginal mucosa showed that DCT gel had no obvious damage to the mucosa epithelium, lamina propria, blood vessels and surrounding tissues (5.5 ± 2.7), which was not significantly different from that of the blank group (3.3 ± 1.6) (P <0.05). Compared with the blank gel group, the expression of CD45 in the vaginal mucosa of DCT gel group was not significantly different (P> 0.05), while the expression of CD45 in N-9 jelly group Significantly increased (P <0.05). Conclusion DCT gel is less irritating to the vaginal mucosa of miniature pigs and has certain application prospects as a vaginal microbiocide.
