目的:探讨热诱导颊癌细胞凋亡过程中Bax基因蛋白及mRNA表达的动态变化,及其在细胞凋亡过程中的作用。方法:43℃水浴40min加热BcaCD885细胞,0,3,6,12,24,48h后收获细胞,采用流式细胞仪DNA蛋白质分析技术、RT-PCR技术检测诱导颊癌细胞凋亡过程中的凋亡率、Bax蛋白及mRNA表达水平。结果:43℃水浴40min后,凋亡率、Bax蛋白及mRNA表达明显升高,与对照组相比,有显著的差异性(P<0.05)。结论:高温作为一种外来刺激信号通过上调bax基因的表达,诱导BcaCD885细胞凋亡的发生。
Objective: To investigate the dynamic changes of Bax gene protein and mRNA expression in heat-induced apoptosis of buccal carcinoma cells and its role in apoptosis. Methods: BcaCD885 cells were heated in water bath at 43 ℃ for 40min, cells were harvested at 0, 3, 6, 12, 24 and 48h. Flow cytometry was used to detect the apoptosis of BcaCD885 cells by RT-PCR. Mortality, Bax protein and mRNA expression levels. Results: The apoptosis rate and the expression of Bax protein and mRNA in the water bath at 43 ℃ for 40min were significantly higher than those in the control group (P <0.05). Conclusion: High temperature, as a kind of external stimulus signal, induces the apoptosis of BcaCD885 cells by up-regulating bax gene expression.
