目的通过对已被GenBank接受、编码与不育有关的人精子蛋白质的cDNA片段(BSD-2.4)的组织特异性及表达阶段的研究,进一步了解其特性。方法采用Dotblot、Northernblot和组织原位杂交技术。结果该cDNA只在人和大鼠睾丸组织中表达;在人睾丸曲细精管的精原细胞、精母细胞和精子细胞中均有转录。结论BSD-2.4cDNA编码蛋白具有睾丸组织特异性,并在精子发生的各个阶段均有不同程度的表达。
OBJECTIVE: To further understand its characteristics through the study of the tissue specificity and expression stage of the cDNA fragment of human sperm protein (GenBank accession, BSD-2.4) encoding for infertility. Methods Dotblot, Northern blot and tissue in situ hybridization were used. Results The cDNA was expressed only in human and rat testes; it was transcribed in spermatogonia, spermatocytes and spermatids in human testicular seminiferous tubules. Conclusion The BSD-2.4 cDNA encodes for testicular tissue specificity and has different degrees of expression in all stages of spermatogenesis.
