目的:研究附睾上皮细胞中功能性TLR2和TLR4的表达情况。方法:采用逆转录聚合酶链式反应(RT-PCR)、Western blot和免疫荧光方法探索原代培养大鼠附睾上皮细胞中TLR2和TLR4的表达情况,同时利用ELISA分别以磷壁酸(LTA)和细菌脂多糖(LPS)对附睾上皮细胞进行功能性检测。结果:TLR2和TLR4在mRNA水平、蛋白水平和细胞水平上在附睾上皮细胞均有表达,TLR2和TLR4的配体LTA和LPS均能激活附睾上皮细胞的天然免疫反应,产生炎症因子TNF-α和IL-1β,差异有统计学意义。结论:附睾上皮细胞表达功能性的TLR2和TLR4。
Objective: To study the expression of functional TLR2 and TLR4 in epididymal epithelial cells. Methods: The expressions of TLR2 and TLR4 in epididymal epithelial cells of primary cultured rats were detected by reverse transcription polymerase chain reaction (RT-PCR), Western blot and immunofluorescence method. And bacterial lipopolysaccharide (LPS) functional test epididymal epithelial cells. Results: Both TLR2 and TLR4 were expressed in epididymal epithelial cells at mRNA level, protein level and cellular level. Both LTA and LPS, both ligands of TLR2 and TLR4, activated innate immune responses of epididymal epithelium and produced inflammatory cytokines TNF-α and IL-1β, the difference was statistically significant. Conclusion: Epididymal epithelial cells express functional TLR2 and TLR4.
