研究中药益气化瘀方对体外培养的大鼠颈椎间盘软骨细胞基质金属蛋白酶mRNA表达的影响。取健康10周龄SD大鼠70只,体重(200±50)g,雌雄各半,随机分为空白组(E)、芬必得组(D)、中药低剂量组(C)、中药中剂量组(B)、中药高剂量组(A),每组14只。定时、定量胃饲,4天后分别处死取血清,相同条件下制备益气化瘀方含药血清和对照血清,取体外培养的大鼠颈椎间盘软骨细胞进行实验研究,采用RT-PCR法。结果显示,各含药血清组培养的软骨细胞均具有分泌MMP-3、TIMP-1功能,但不具有诱导其异常分泌MMP-3、TIMP-1功能。益气化瘀方组较对照组及芬必得组有下调MMP-3的分泌或表达,上调TIMP-1的分泌或表达。
To study the effect of Yiqi Huayu Recipe on the expression of matrix metalloproteinase mRNA in cultured rat cervical intervertebral disc chondrocytes. 70 healthy 10-week-old Sprague-Dawley rats were weighed (200±50) g and divided into blank group (E), fenbide group (D), low dose group of traditional Chinese medicine (C), and medium dose of Chinese medicine. Group (B), high dose group (A), 14 in each group. Timely and quantitative gastro- feeding were performed. Four days later, the serum was sacrificed. Under the same conditions, Yiqi Huayufang-containing serum and control serum were prepared. The rat cervical disc chondrocytes cultured in vitro were subjected to experimental research and RT-PCR was used. The results showed that chondrocytes cultured in each serum containing sera had the functions of secreting MMP-3 and TIMP-1, but did not induce the abnormal secretion of MMP-3 and TIMP-1 functions. The Yiqi Huayu Recipe group had down-regulated the secretion or expression of MMP-3 and increased the secretion or expression of TIMP-1 compared with the control group and the Fenbid group.
