目的 :探讨缝隙连接蛋白 43 (Cx43 )和E 钙黏素 (E cad)在口腔鳞癌组织中的表达及其生物学行为的关系。方法 :SABC法对 8例正常口腔鳞状上皮 ,2 9例不同分化程度的口腔鳞癌 (包括舌癌 )石蜡切片进行Cx43和E cad表达的半定量检测 ;同时应用免疫细胞化学对舌癌细胞株 (Tca8113 )Cx43和E Cad的表达进行检测。结果 :Cx43和E cad在正常口腔鳞状上皮主要表达于细胞膜上 ,Cx43和E Cad表达的阳性面积和灰度值在正常口腔鳞状上皮与高分化鳞癌之间无明显差异 (P >0 .0 5 )。但随着病理分级的升高 ,两指标的下降具有统计学意义 (P <0 .0 5 )。在口腔鳞癌组织中 ,Cx43和E cad的表达与组织的分化程度、转移之间有显著性差异 (Cx43 :χ2 =7.42、12 .43 ,P <0 .0 5、P <0 .0 1;E cad :χ2 =7.79、9.688,P <0 .0 5、P <0 .0 1)。同一标本中Cx43和E cad表达具有正相关性和一致性 (r =0 .5 77,P <0 .0 0 0 5 )。结论 :Cx43和E Cad的表达水平可作为口腔鳞癌分化和转移的生物学标志。
Objective: To investigate the relationship between the expression of connexin 43 (Cx43) and E cadherin in oral squamous cell carcinoma and their biological behavior. Methods: SABC method was used to detect the expression of Cx43 and E cad in 8 cases of normal oral squamous epithelium and 29 cases of oral squamous cell carcinoma (including tongue cancer) with different degrees of differentiation. Meanwhile, immunocytochemistry was used to detect the expression of Cx43 and E cad in tongue squamous cell carcinoma Strains (Tca8113) Cx43 and E Cad expression were detected. Results: The positive expression of Cx43 and E cad in normal oral squamous epithelium was mainly expressed on the cell membrane. The positive area and gray value of Cx43 and E Cad expression were not significantly different between normal oral squamous cell carcinoma and well differentiated squamous cell carcinoma (P> 0.05) .0 5). However, with the increase of pathological grade, the decline of the two indexes was statistically significant (P <0.05). In oral squamous cell carcinoma, there was a significant difference between the expression of Cx43 and E cad and histological differentiation and metastasis (Cx43: χ2 = 7.42,12.43, P <0.05, P <0.01 ; E cad: χ2 = 7.79,9.688, P <0.05, P <0.01). The same specimen Cx43 and E cad expression has a positive correlation and consistency (r = 0.5577, P <0.00005). Conclusion: The expression levels of Cx43 and E Cad can be used as biomarkers for the differentiation and metastasis of oral squamous cell carcinoma.
