目的:建立粟米草(Mollugo pentaphylla L.)的毛状根培养系统并研究毛状根的生长特性。方法:分别用发根农杆菌R1601,ATCC 15834和A4感染粟米草无菌苗的子叶,获得毛状根,并筛选优质株系;用高压纸电泳法对毛状根进行甘露碱检测;测定其生长曲线;比较不同基本培养液、不同pH 值对毛状根生长的影响。结果:高压纸电泳结果显示,所获毛状根中均含有甘露碱,表明毛状根中确已转入发根农杆菌的T-DNA;1/2 MS0培养液pH 5.4~5.8最适合粟米草毛状根的生长。结论:快速生长的粟米草毛状根在药用活性成分的生产上将具有较大潜力。
Objective: To establish the hairy root culture system of Mollugo pentaphylla L. and study the growth characteristics of hairy roots. Methods: The cotyledon of A. rhizogenes R1601, ATCC 15834 and A4 were used to infect the cotyledon of corn seedlings, and the hairy roots were obtained. High quality strains were selected. The hairy roots were detected by high pressure paper electrophoresis. Growth curve; compare the effects of different basic culture fluids and different pH values on hairy root growth. RESULTS: High-pressure paper electrophoresis showed that the hairy roots contained mannopine, indicating that the hairy root had indeed transferred to T-DNA of Agrobacterium rhizogenes; 1/2 MS0 medium pH was 5.4 to 5. 8 is most suitable for the growth of corn grassy hairy roots. Conclusion: The rapidly growing hairy roots of corn grass will have great potential in the production of medicinal active ingredients.
