利用植物微管特异解聚剂APM(Amiprophos-methyl)处理小麦幼苗,会导致其植株矮小,根尖伸长受阻并变粗膨大,尤以分生区部分膨大最为明显。光镜观察发现:APM处理的小麦根尖,其根冠、分生区、伸长区细胞均有不同程度的膨大变形,分生区细胞分裂消失。电镜观察发现:经APM处理的小麦根尖分生区细胞内液泡明显增多,质体数目明显减少,核膜也变得凹凸不平。研究表明,微管与小麦根尖细胞形状、大小及分生区细胞器的结构、数量均有着一定的关系。
The treatment of wheat seedlings with plant microtubule-specific depolymerizing agent APM (Amiprophos-methyl) will result in short plants, shortened root apical elongation and thickening, especially in the meristematic area. Light microscopy found: APM-treated wheat root tip, the root crown, meristematic zone, elongation zone cells have varying degrees of deformation, meristematic cell division disappeared. Electron microscopy showed that the number of vacuoles in the apical meristematic area of APM-treated wheat significantly increased, the number of plastids decreased, and the nuclear membrane also became uneven. Studies have shown that the microtubules and wheat root tip cells in shape, size and meristematic organelle structure and quantity have a certain relationship.
