在确证自发性高血压大鼠(SHR)模型成功后比较其与正常大鼠(Wistar)尿液氨基酸代谢谱的差异,在此基础上初步研究两色金鸡菊提取物对SHR血压及氨基酸代谢谱的调节作用。选取经过适应性饲养后的适龄雄性SHR大鼠和同龄Wistar大鼠测量大鼠收缩压(SBP),舒张压(DBP),同时收集大鼠尿液,运用AQC柱前衍生高效液相色谱(HPLC)-荧光法测定尿液中氨基酸代谢谱,利用多元统计分析中的偏最小二乘方判别分析(PLS-DA)研究2组动物的氨基酸代谢差异物。将所有饲养的SHR大鼠随机分成5组,即模型组,两色金鸡菊提取物高、中、低剂量组(3.2,1.6,0.8 g·kg~(-1)),西药卡托普利组(4mg·kg~(-1)),连续灌胃给药4周后收集大鼠尿液,测定各组氨基酸代谢谱。药物干预4周后,与Wistar组相比,SHR组的氨基酸代谢谱中丝氨酸、丙氨酸、酪氨酸、胱氨酸对分组的贡献较大,均显著高于Wistar组;与SHR组相比,卡托普利组的苏氨酸、蛋氨酸较模型组显著降低(P<0.01);新疆两色金鸡菊醇提物低、中、高剂量组氨基酸代谢均有不同程度改变,其中低剂量苏氨酸较模型组显著降低(P<0.01);中剂量组的丝氨酸、苏氨酸较模型组降低(P<0.05),缬氨酸、蛋氨酸、赖氨酸显著降低(P<0.01);大剂量组的苏氨酸、缬氨酸、蛋氨酸、赖氨酸显著降低(P<0.01)。研究结果显示,中、高剂量提取物能明显改善氨基酸代谢异常,帮助进一步阐释两色金鸡菊的药性研究,为高血压病存在氨基酸代谢通路异常提供数据支持。

The amino acid metabolism of alcoholic extract of Bismillanticolum in Xinjiang under low, medium and high dose groups changed to different degrees, Threonine was significantly lower than the model group (P <0.01). Serine and threonine of middle dose group were lower than those of the model group (P <0.05), and valine, methionine and lysine were significantly decreased (P <0.01); Threonine, valine, methionine and lysine were significantly decreased in the high-dose group (P <0.01). The results showed that medium and high dose of extract can significantly improve the abnormalities of amino acid metabolism and help further explain the drug resistance of two color Coreopsis, and provide data support for the existence of abnormal amino acid metabolism pathways in hypertension.
