目的观察糖皮质激素对大鼠股骨的影响。方法4个月龄雌性SD大白鼠。随机分为3组,A组为正常对照组(生理盐水,5ml/(kg.d)灌胃);B组为低剂量激素组(泼尼松2ml/(kg.d)灌胃);C组为高剂量激素组(泼尼松6ml/(kg.d)灌胃)。1次/d,连续18周。实验结束时,处死全部大鼠,取股骨上端进行不脱钙骨制片,用计算机全自动图像分析系统进行骨组织学形态计量学分析。结果与A组相比,B组各项指标无明显下降;C组的TbAr、Tb.N、Tb.Th明显减少,Tb.Sp明显增加,说明C组大鼠骨量明显减少。结论长期使用高剂量糖皮质激素,可以引起大鼠骨量减少,发生骨质疏松。长期使用低剂量糖皮质激素不引起大鼠骨质疏松。大鼠股骨颈是研究骨质疏松模型和药物对此模型作用的重要部位。
Objective To observe the effect of glucocorticoids on the femur of rats. Methods 4-month-old female SD rats. A group was normal control group (saline, 5ml / (kg.d) intragastrically); B group was low dose hormone group (prednisone 2ml / (kg.d) intragastric administration); C Group for the high-dose hormone group (prednisone 6ml / (kg.d) gavage). 1 times / d, for 18 weeks. At the end of the experiment, all the rats were sacrificed, and the upper part of the femur was taken for non-decalcified bone preparation. Bone histomorphometry was analyzed by computer automatic image analysis system. Results Compared with group A, the indexes of group B did not decrease significantly. The levels of TbAr, Tb.N, Tb.Th and group C significantly increased in group C, which indicated that the bone mass of group C decreased significantly. Conclusion Long-term use of high-dose glucocorticoids can cause osteopenia and osteoporosis in rats. Long-term use of low-dose glucocorticoid does not cause osteoporosis in rats. The femoral neck of rats is an important part of studying the effects of osteoporosis models and drugs on this model.
