目的:建立Aurora-A高表达的稳定细胞系,并探讨Aurora-A高表达对食管癌细胞侵袭和MMP-9蛋白表达与活性的影响。方法:利用脂质体法稳定转染Aurora-A表达质粒,通过Westernblot和RT-PCR鉴定Aurora-A高表达的稳定细胞系。利用细胞体外侵袭实验观察细胞的侵袭能力,并通过Western blot、ELISA和明胶酶谱的方法观察MMP-9的蛋白表达水平和活性。结果:建立了Aurora-A高表达的稳定细胞系,且Aurora-A高表达能增加食管癌细胞的体外侵袭能力,并提高了MMP-9的蛋白表达水平和活性。结论:初步阐明了Aurora-A促进食管癌细胞侵袭的作用以及分子机制,为Aurora-A可能成为治疗食管癌的有效靶点提供了依据。
OBJECTIVE: To establish a stable Aurora-A expressing cell line and investigate the effect of Aurora-A overexpression on esophageal cancer cell invasion and MMP-9 protein expression and activity. Methods: Aurora-A expression plasmid was stably transfected by liposome and the stable cell line highly expressing Aurora-A was identified by Western blot and RT-PCR. The invasion ability of cells was observed by in vitro invasion assay. The protein expression and activity of MMP-9 were observed by Western blot, ELISA and gelatin zymography. RESULTS: Aurora-A highly stable cell line was established. Aurora-A overexpression enhanced the invasiveness of esophageal carcinoma cells in vitro and increased the protein expression and activity of MMP-9. CONCLUSION: The preliminary elucidation of the role of Aurora-A in esophageal cancer cell invasion and its molecular mechanism provide a basis for Aurora-A to be an effective target for the treatment of esophageal cancer.
