目的探讨延肾1号冲剂抗肾缺血再灌注损伤的作用机理。方法将72只大鼠随机分为假手术组、对照组、治疗组,各24只。治疗组给予延肾1号冲剂,假手术组及对照组灌胃相应量的自来水。检测大鼠肾缺血1h,再灌注24、48h后肾功能(BUN、Cr)的变化及肾组织肿瘤坏死因子(TNF-α)和细胞间黏附因子(ICAM-1)的变化情况。结果假手术组大鼠肾小管上皮细胞内可见微量TNF-α、ICAM-1表达。与假手术组相比,治疗组、对照组TNF-α、ICAM-1分子表达增强,在肾缺血再灌注24、48h有显著差异(P<0.01);与对照组比较,治疗组TNF-α、ICAM-1分子表达下调,肾缺血1h无显著差异(P>0.05),再灌注24、48h有显著差异(P<0.01)。结论延肾1号冲剂能够保护肾缺血再灌注大鼠的肾功能并抑制其肾组织中TNF-α、ICAM-1的蛋白表达。
Objective To investigate the mechanism of action of Yanshen No. 1 Granule on renal ischemia reperfusion injury. Methods 72 rats were randomly divided into sham operation group, control group and treatment group, 24 rats in each group. The treatment group was given Yanshen No. 1 granules, and the sham operation group and the control group were given the corresponding amount of tap water. The changes of renal function (BUN, Cr) and the changes of tumor necrosis factor-α (TNF-α) and intercellular adhesion molecule (ICAM-1) in renal tissue were detected at 1 hour after renal ischemia in rats, 24 and 48 hours after reperfusion. Results The expression of TNF-α and ICAM-1 in renal tubular epithelial cells was observed in sham-operated rats. Compared with the sham-operated group, the expression of TNF-α and ICAM-1 in the treatment group and the control group increased, and there was a significant difference in renal ischemia-reperfusion 24 and 48 hours (P<0.01). Compared with the control group, the treatment group TNF- The expression of α and ICAM-1 molecules was down-regulated, and there was no significant difference in renal ischemia at 1 hour (P>0.05). There was a significant difference between 24 and 48 hours after reperfusion (P<0.01). Conclusion Yanshen No. 1 Granule can protect the renal function of renal ischemia-reperfusion rats and inhibit the expression of TNF-α and ICAM-1 in renal tissue.
