目的探讨姜黄素对急性心肌梗死小鼠心肌细胞凋亡相关基因表达的影响。方法 30只健康昆明种小鼠随机分为假手术组、模型组和姜黄素组。假手术组腹腔注射9 g.L-1盐水10 mL.kg-1,每隔30 min注射1次,共2次;模型组腹腔注射9 g.L-1盐水10 mL.kg-1,30 min后腹腔注射垂体后叶素(Pit)10 mL.kg-1;姜黄素组腹腔注射姜黄素100 mg.kg-1,30 min后腹腔注射Pit10 mL.kg-1。ELISA法检测各组小鼠血清乳酸脱氢酶(LDH)和心肌肌钙蛋白I(cTnI)水平。采用RT-PCR法检测各组小鼠心肌组织Fas mRNA及FasLmRNA的表达水平。结果与假手术组相比,模型组血清LDH、cTnI和心肌组织Fas mRNA及FasL mRNA表达水平显著升高(Pa<0.01);与模型组相比,姜黄素组血清LDH、cTnI和心肌组织Fas mRNA及FasLmRNA的表达水平显著降低(Pa<0.01)。结论姜黄素对急性心肌梗死小鼠有较好的保护作用,其机制可能与下调Fas mRNA及FasLmRNA的表达有关。
Objective To investigate the effect of curcumin on expression of cardiomyocyte apoptosis-related genes in mice with acute myocardial infarction. Methods 30 healthy Kunming mice were randomly divided into sham operation group, model group and curcumin group. The sham group was injected intraperitoneally with 9 gL-1 saline 10 mL.kg-1 and injected once every 30 minutes for 2 times. The model group was injected with 9 gL-1 saline 10 mL.kg-1 by intraperitoneal injection and injected intraperitoneally 30 minutes later. Pituitrin (Pit) was 10 mL.kg-1; curcumin group was injected intraperitoneally with curcumin 100 mg.kg-1, and 30 min later, Pit 10 mL.kg-1 was injected intraperitoneally. The serum levels of lactate dehydrogenase (LDH) and cardiac troponin I (cTnI) were measured by ELISA. The expression of Fas mRNA and FasL mRNA in myocardium were detected by RT-PCR. Results Compared with the sham operation group, serum LDH, cTnI, and FasL mRNA expression levels in the model group were significantly increased (Pa<0.01). Compared with the model group, serum LDH, cTnI, and myocardial tissue Fas in the curcumin group The expression levels of mRNA and FasL mRNA were significantly decreased (Pa<0.01). Conclusion Curcumin can protect mice with acute myocardial infarction. The mechanism may be related to the down-regulation of Fas mRNA and FasL mRNA expression.
