目的:对滇白珠的各提取液进行抗大鼠佐剂性关节炎活性筛选,为临床应用滇白珠治疗风湿性关节炎提供实验基础。方法:制备滇白珠的水提取总液、乙醇提取总液、醋酸乙酯提取液、正丁醇提取液、乙醇提取总液剩余部分液。滇白珠的正丁醇部分,在大孔树脂中用不同溶液洗脱,得到水洗脱部分、30%乙醇洗脱部分和95%乙醇洗脱部分。采用大鼠佐剂性关节炎模型,观察滇白珠不同部分提取液对大鼠关节肿胀的影响。结果:滇白珠的水提液组(A)、酸酸乙酯提取液组(C)和正丁醇提取液组(D)可明显减轻大鼠关节肿胀程度,正丁醇提取液组(D)作用最强(P<0.05,P<0.01)。进一步研究显示,滇白珠正丁醇提取液中水洗脱液和30%乙醇洗脱液均有抗大鼠佐剂性关节炎作用,而30%乙醇洗脱液作用更明显(P<0.05,P<0.01)。结论:滇白珠正丁醇提取液30%乙醇洗脱部分有明显的抗大鼠佐剂性关节炎作用。
OBJECTIVE: To screen anti-rats’ adjuvant arthritis in each extract of Radix Paeoniae Alba, and provide experimental basis for clinical application of Radix Paeoniae Alba for rheumatoid arthritis. Methods: The total aqueous solution of Eriobotrya japonica, the total ethanol extraction solution, the ethyl acetate extraction solution, the n-butanol extraction solution, and the remaining portion of the ethanol extraction total solution were prepared. The n-butanol portion of the beaded white beads was eluted with a different solution in a macroporous resin to obtain a water eluted fraction, a 30% ethanol elution fraction, and a 95% ethanol elution fraction. Rat adjuvant arthritis model was used to observe the effects of extracts of different parts of Radix Paeoniae Alba on joint swelling of rats. RESULTS: The water extract group (A), ethyl acid ethyl acetate extract group (C) and n-butanol extract group (D) of Radix Paeoniae Alba can significantly reduce the degree of joint swelling in rats, n-butanol extract group (D The strongest effect was (P<0.05, P<0.01). Further studies showed that the water eluent and 30% ethanol eluent in the n-butanol extract of Radix Paeoniae Alba had anti-rat arthritis effect, but the effect of 30% ethanol eluent was more pronounced (P<0.05). P<0.01). Conclusion: The fraction eluted by 30% ethanol from the extract of Nongbaizhu n-butanol has obvious anti-rat adjuvant arthritis effect.
