目的:探讨鼻咽癌细胞中细胞周期蛋白G1(cyclin G1)、细胞周期蛋白G2(cyclin G2)基因的表达及其意义。方法:利用细胞培养技术、免疫荧光技术检测并分析不同分化程度的鼻咽癌细胞株HNE2(B010)、CNE1(B003)中cyclinG1、cyclin G2的表达情况,逆转录聚合酶链反应技术(RT-RCR)检测HNE2、CNE1中cyclin G1-mRNA、cyclin G2-mR-NA的表达水平。结果:cyclin G1蛋白在HNE2、CNE1呈现弱阳性表达,染色较弱,cyclin G2蛋白在HNE2、CNE1呈现强阳性表达,在靠近细胞膜的细胞质中染色较强,连续且均匀一致。HNE2中cyclin G1mRNA的表达水平(2.097±0.262)明显高于CNE1(0.997±0.286,P<0.01),HNE2中cyclin G2mRNA的表达水平(0.708±0.107)明显低于CNE1(1.216±0.037,P<0.01)。结论:细胞周期素G表达水平与鼻咽癌分化程度密切相关,可作为鼻咽癌恶性程度及发生、发展的指标之一。
Objective: To investigate the expression of cyclin G1 and cyclin G2 in nasopharyngeal carcinoma cells and its significance. Methods: The expression of cyclinG1 and cyclin G2 in nasopharyngeal carcinoma cell line HNE2 (B010) and CNE1 (B003) with different differentiation degrees were detected and analyzed by immunofluorescence technique using cell culture technique and RT- RCR) was used to detect the expression of cyclin G1-mRNA and cyclin G2-mR-NA in HNE2 and CNE1. Results: The expression of cyclin G1 protein in HNE2 and CNE1 cells was weakly positive. The expression of cyclin G2 protein was strongly positive in HNE2 and CNE1 cells. The staining of cyclin G1 protein in the cytoplasm was close and uniform. The expression level of cyclin G1 mRNA in HNE2 was significantly higher than that in CNE1 (2.097 ± 0.262, 0.997 ± 0.286, P <0.01). The expression of cyclin G2 mRNA in HNE2 was significantly lower than that of CNE1 (1.216 ± 0.037, P <0.01) ). Conclusion: The expression of cyclin G is closely related to the degree of differentiation of nasopharyngeal carcinoma and may be used as one of the indicators of malignant degree, occurrence and development of nasopharyngeal carcinoma.
