目的探讨M2型巨噬细胞在食管癌细胞增殖、迁移、侵袭和上皮间质转化(epithelial-mesenchymal transition,EMT)中的作用。方法将人急性白血病单核细胞(THP-1)诱导分化为M2型巨噬细胞,然后将其与食管鳞癌Eca109和KYSE-150细胞系共培养,通过细胞增殖(MTT)、细胞划痕及细胞侵袭实验分别评价共培养细胞(共培养组)与非共培养细胞(非共培养组)增殖、迁移和侵袭能力的变化,并运用Western-blot检测EMT相关指标E-钙黏蛋白(E-cadherin)、波形蛋白(Vimentin)和锌指转录因子(Slug)的表达变化。结果与非共培养组相比,共培养组食管鳞癌细胞迁移和侵袭能力明显增强(P<0.05),但其增殖能力差异无统计学意义(P>0.05)。Western-blot检测显示,上皮标志物E-cadherin表达减少,间质标志物Vimentin和Slug表达增加。结论 M2型巨噬细胞促进食管鳞癌细胞的迁移、浸润及上皮间质转化,但对其增殖无明显影响。
Objective To investigate the role of M2 macrophages in the proliferation, migration, invasion and epithelial-mesenchymal transition (EMT) of esophageal cancer cells. Methods Human acute leukemia monocyte (THP-1) was induced to differentiate into M2 type macrophages and then co-cultured with esophageal squamous cell carcinoma Eca109 and KYSE-150 cell lines. MTT, cell scratch, The cell invasion assay evaluated the proliferation, migration and invasion ability of co-cultured cells (co-culture group) and non-co-cultured cells (non-co-culture group) respectively. Western blot analysis was used to detect the expression of EMT-related marker E-cadherin cadherin, vimentin and zinc finger transcription factor (Slug) expression changes. Results Compared with the non-co-culture group, the ability of migration and invasion of esophageal squamous carcinoma cells in co-culture group was significantly enhanced (P <0.05), but the proliferation ability was not significantly different (P> 0.05). Western-blot showed that the expression of epithelial marker E-cadherin decreased and the expression of interstitial markers Vimentin and Slug increased. Conclusion M2 macrophages promote esophageal squamous cell carcinoma cell migration, invasion and epithelial-mesenchymal transition, but have no significant effect on its proliferation.
