目的检测用9-cis-RA处理前后肺腺癌细胞株PG和A549维甲酸受体RARγ不同时相的表达变化。方法将两细胞株分别分为9-cis-RA组和对照组,应用细胞计数方法统计每组各时相细胞数,相对定量RT-PCR方法检测加药前后RARγ在不同时相的表达变化。结果加入9-cis-RA前,PG和A549细胞RARγmRNA转录水平检测结果显示RARγ在PG细胞株中有一定水平的基础转录,而在A549细胞中无基础转录。应用1uM9-cis-RA后,PG细胞的RARγmRNA的转录水平受到抑制,而A549细胞的RARγ的转录水平在各时相有较弱的诱导表达。结论RARγ受体对9-cis-RA的诱导在不同细胞株中的反应不同,表明9-cis-RA对不同肿瘤细胞株的作用存在特异性。
Objective To detect the expression changes of RARγ in PGCs and A549 cells before and after treatment with 9-cis-RA. Methods The two cell lines were divided into 9-cis-RA group and control group. Cell counting method was used to count the number of cells in each phase. Relative quantitative RT-PCR was used to detect the expression of RARγ at different time points. Results Before transfection with 9-cis-RA, the transcriptional level of RARγ mRNA in PG and A549 cells showed that RARγ had some basal transcription in PG cells but no basic transcription in A549 cells. After 1uM9-cis-RA was applied, the transcriptional level of RARγmRNA in PG cells was inhibited, while the transcriptional level of RARγ in A549 cells was weakly induced at each phase. Conclusion The RARγ receptor has different responses to 9-cis-RA in different cell lines, indicating that 9-cis-RA has specific effects on different tumor cell lines.
