从忍冬(Lonicera japonica Thunb.)转录组测序结果中分析获得1个FatB基因。分别以忍冬、红白忍冬(Lonicera japonica Thunb.var.chinensis(Wats.)Bak.)、红腺忍冬(Lonicera hypoglauca Miq.)和水忍冬(Lonicera dasystyla Rehd.)新鲜花蕾为材料,利用RT-PCR技术克隆获得了LJFatB、LHFatB、LJCFatB、LDFatB基因的全长cDNA,并进行生物信息学分析。结果表明LJFatB、LJCFatB、LHFatB和LDFatB与拟南芥AtFatB具有较近的亲缘关系。LJFatB、LJCFatB、LHFatB和LDFatB的核苷酸序列、蛋白特性及其二级结构有所差异,但其蛋白均具有保守的FatB底物结合位点和催化活性位点,且在花蕾中的转录水平没有显著差异。因此推测LJFatB、LJCFatB、LHFatB和LDFatB可能具有与AtFatB相同的生物学功能。
One FatB gene was obtained from the sequencing results of transcriptome of Lonicera japonica Thunb. The fresh buds of Lonicera japonica Thunb.var.chinensis (Wats.) Bak., Lonicera hypoglaucca Miq. And Lonicera dasystyla Rehd. The full-length cDNA of LJFatB, LHFatB, LJCFatB and LDFatB was cloned by bioinformatics analysis. The results showed that LJFatB, LJCFatB, LHFatB and LDFatB have a close genetic relationship with Arabidopsis AtFatB. The nucleotide sequence, protein characteristic and secondary structure of LJFatB, LJCFatB, LHFatB and LDFatB are different, but all of them have conserved FatB substrate binding sites and catalytic active sites, and the transcriptional levels in buds No significant difference. Therefore, it is speculated that LJFatB, LJCFatB, LHFatB and LDFatB may have the same biological function as AtFatB.
