目的探讨乙酰肝素酶2(Hpa2)mRNA和蛋白在胰腺癌中的表达及其意义。方法应用逆转录-聚合酶链反应(RT-PCR)及免疫组织化学染色法检测54例胰腺癌组织中Hpa2 mRNA和蛋白的表达。结果正常胰腺组织中Hpa2 mRNA及蛋白均未检出,胰腺癌组织中Hpa2 mRNA的表达及患者的年龄(<60岁20/27:≥60岁24/27)、性别(男26/30:女18/24)、肿瘤的直径(<2 cm 15/20:≥2 cm 29/34)均无关(P>0.05),在临床分期高的肿瘤(35/39,89.74%)中的表达明显高于分期低(9/15,60.00%),包膜完整(6/12,50.00%),临近组织无浸润(7/12,58.33%)和无远处淋巴结转移(4/10,40.00%)者(P<0.05)。Hpa2蛋白的表达与患者的年龄(<60岁21/27:≥60岁22/27)、性别(男23/30:女20/24)、肿瘤的直径(<2 cm 17/20:≥2 cm 26/34)均无关(P>0.05),而Hpa2蛋白在临床分期高(35/39,89.74%),包膜不完整(37/42,88.09%),浸润临近组织(38/42,90.48%),有淋巴结转移(39/44,88.64%)的肿瘤中的表达明显高于分期低(8/15,53.33%),包膜完整(6/12,50.00%),临近组织无浸润(5/12,41.66%)和无淋巴结转移(8/26,30.77%)肿瘤中的表达(P<0.05)。结论Hpa2 mRNA和蛋白与肿瘤的临床分期、包膜完整与否、浸润临近组织、远处淋巴结转移有关,可以作为判断侵袭、转移的指标。
Objective To investigate the expression of heparanase 2 (Hpa2) mRNA and protein in pancreatic carcinoma and its significance. Methods The expression of Hpa2 mRNA and protein in 54 pancreatic cancer tissues was detected by reverse transcription polymerase chain reaction (RT-PCR) and immunohistochemical staining. Results Hpa2 mRNA and protein were not detected in normal pancreatic tissues. The expression of Hpa2 mRNA in pancreatic cancer tissues and the age of the patients (<60 years old and 20/27: ≥ 60 years old were 24/27) 18/24), tumor diameter (<2 cm 15/20: ≥2 cm 29/34) (P> 0.05), and significantly higher in tumors with high clinical stage (35/39, 89.74%) (7/12, 58.33%) and no distant lymph node metastasis (4/10, 40.00%) in the advanced stage (9/15, 60.00% (P <0.05). The expression of Hpa2 protein correlated with the age of patients (<60 years old 21/27: ≥60 years old 22/27), sex (male 23/30: female 20/24), tumor diameter (<2 cm 17/20: cm 26/34), while the expression of Hpa2 protein in clinical stage (35 / 39,89.74%), incomplete capsule (37 / 42,98.09%), infiltration adjacent tissue (38/42, 90.48%). The expression of tumor with lymph node metastasis (39/44, 88.64%) was significantly lower than that of stage (8/15, 53.33%), complete capsule (6/12, 50.00% (5 / 12,41.66%) and no lymph node metastasis (8 / 26,30.77%) tumors (P <0.05). Conclusion The expression of Hpa2 mRNA and protein may be related to the clinical stage, integrity of the capsule, adjacent infiltration, and distant lymph node metastasis, which may be used as an indicator of invasion and metastasis.
