目的:探讨环氧化酶-2(COX-2)在大鼠动脉血栓中的表达及意义。方法:利用氯化铁溶液外敷大鼠左颈总动脉形成动脉血栓,HE染色后观察不同外敷时间组(1 d、3 d、7 d、14 d)大鼠的左颈总动脉血栓镜下的结构特点,采用免疫组化染色分析血栓中COX-2蛋白的表达情况,用蛋白免疫法分析不同时间组大鼠动脉血栓及循环血白细胞总蛋白,观察不同时间组动脉血栓标本及白细胞中COX-2的表达情况。结果:与正常对照组比较,外敷氯化铁溶液后,实验组大鼠的动脉可见明显的血栓形成。免疫组化染色显示,对照组未中见COX-2蛋白阳性表达,而实验组中14 d组血栓中可见多处COX-2蛋白阳性染色。蛋白印迹检测显示,对照组大鼠的COX-2仅有极少量表达,但均明显低于实验组(P<0.01),而实验组造模1 d后血栓中COX-2的表达量有所升高,3 d后表达量出现明显上升,7 d后表达量继续升高,造模14 d后,其含量进一步升高。血栓内的炎性细胞主要成分在第1天和第3天以中性粒细胞为主,而第7天和第14天则以单核巨噬细胞为主。结论:COX-2蛋白表达在大鼠颈动脉血栓中明显增加,提示COX-2的表达与动脉血栓形成有关,可能参与了动脉血栓形成的进程。
Objective: To investigate the expression of cyclooxygenase-2 (COX-2) in rat arterial thrombosis and its significance. Methods: Arterial thrombosis was induced by application of ferric chloride solution in left common carotid arteries of rats. HE staining was used to observe the changes in left common carotid artery thrombectomy in rats with different external application time (1 d, 3 d, 7 d, 14 d) The protein expression of COX-2 in thrombus was analyzed by immunohistochemical staining. The contents of COX-2 protein in arterial thrombus and circulating white blood cells in different time groups were analyzed by protein immunoassay. 2 of the expression. Results: Compared with the normal control group, obvious external thrombosis was observed in the arteries of rats in experimental group after application of ferric chloride solution. Immunohistochemical staining showed that the COX-2 protein expression was not observed in the control group, while COX-2 protein positive staining was found in the thrombus in the 14 d group in the experimental group. Western blotting showed that COX-2 expression was only slightly decreased in the control group (all P <0.01), while the COX-2 expression in the experimental group was significantly lower than that in the experimental group After 3 days, the expression increased obviously. After 7 days, the expression continued to increase. After 14 days, the content increased further. The main components of inflammatory cells in the thrombus are mainly neutrophils on day 1 and day 3, and monocytes on day 7 and 14. CONCLUSION: COX-2 protein expression is significantly increased in carotid artery thrombosis rats, suggesting that the expression of COX-2 is associated with arterial thrombosis and may be involved in the process of arterial thrombosis.
