目的探讨高糖状态下胰岛素样生长因子1(IGF-1)对体外培养的大鼠雪旺细胞neuritinmRNA及蛋白表达的影响。方法体外培养的大鼠雪旺细胞分为五组:A组为非干预对照组;B、C、D和E组分别以浓度为0.1、1、10和100ng/ml的IGF-1并高糖同步处理36h。用实时荧光定量PCR(RT-qPCR)、蛋白免疫印迹实验分别检测大鼠雪旺细胞neuritin mRNA和蛋白的表达量。结果B、C组雪旺细胞neuritin mRNA和蛋白表达量与A组相仿(P>0.05),D、E组雪旺细胞neuritinmRNA和蛋白表达量均显著高于A组(P<0.05)。D组与E组雪旺细胞neuritin mRNA和蛋白表达量比较无统计学差异(P>0.05)。结论 IGF-1能上调体外高糖培养的大鼠雪旺细胞neuritinmRNA及蛋白的表达量。
Objective To investigate the effect of insulin-like growth factor-1 (IGF-1) on neuritin mRNA and protein expression in rat Schwann cells cultured in vitro with high glucose. Methods Rat Schwann cells cultured in vitro were divided into five groups: group A was non-intervention control group; group B, C, D and E were treated with IGF-1 at concentrations of 0.1, 1, 10 and 100 ng / ml, Synchronization 36h. Real-time quantitative PCR (RT-qPCR) and Western blotting were used to detect the expression of neuritin mRNA and protein in rat Schwann cells. Results The expressions of neuritin mRNA and protein in Schwann cells in groups B and C were similar to those in group A (P> 0.05). The expressions of neuritin mRNA and protein in Schwann cells in groups D and E were significantly higher than those in group A (P <0.05). There was no significant difference in neuritin mRNA and protein expression between Schwann cells in D group and E group (P> 0.05). Conclusion IGF-1 can up-regulate the expression of neuritin mRNA and protein in rat Schwann cells cultured in vitro with high glucose.
