目的探讨辛芷鼻敏胶囊对变应性鼻炎大鼠的血清中白细胞介素-5(IL-5)、白细胞介素-13(IL-13)和鼻黏膜组织中的eotaxin、CCR3、信号转导和转录激活因子-6(STAT6)基因表达的影响,探讨其可能的作用机制。方法采用卵白蛋白制备变应性鼻炎大鼠模型。造模后大鼠随机分为模型组、辛芷鼻敏胶囊(2.3 g/kg)组、鼻炎康(1.8 g/kg)阳性对照组,每天给药1次,连续给药10 d,另设对照组。ELISA法检测各组大鼠血清IL-5、IL-13水平;用原位杂交(ISH)法半定量检测鼻黏膜中eotaxin、CCR3、STAT6基因的表达。结果辛芷鼻敏胶囊组大鼠血清IL-5、IL-13水平和鼻黏膜中STAT6基因表达明显低于模型组及鼻炎康组(P<0.05)。结论辛芷鼻敏胶囊可能通过下调STAT6基因表达,抑制大鼠变应性鼻炎发病过程中IL-5、IL-13的水平,达到减少嗜酸性粒细胞浸润进而改善变应性鼻炎症状的目的。
Objective To investigate the effects of Xinzhi bimin capsule on serum levels of interleukin-5 (IL-5), interleukin-13 (IL-13) and nasal mucosa in rats with allergic rhinitis And activator of transcription-6 (STAT6) gene expression, explore its possible mechanism of action. Methods Ovalbumin was used to prepare rat model of allergic rhinitis. The rats were randomly divided into model group, Xinzhi bimin capsule (2.3 g / kg) group and rhinitis and Kangyan (1.8 g / kg) positive control group, once a day for continuous administration for 10 days. Control group. Serum levels of IL-5 and IL-13 were detected by ELISA. The expression of eotaxin, CCR3 and STAT6 in nasal mucosa were detected by semi-quantitative in situ hybridization (ISH). Results Serum levels of IL-5 and IL-13 and STAT6 gene expression in nasal mucosa of Xinzhi bimin capsule group were significantly lower than those in model group and rhinitis group (P <0.05). Conclusion Xinzhi bimin capsule may reduce the eosinophil infiltration and improve the symptoms of allergic rhinitis by down-regulating STAT6 gene expression and inhibiting the levels of IL-5 and IL-13 during the pathogenesis of allergic rhinitis in rats.
