目的探讨体外成骨诱导的兔骨髓间充质干细胞(BMSCs)的形态和功能特征。方法体外培养获取生长状态良好的兔BMSCs,进行成骨诱导,观察并检测成骨细胞的生物学特性。结果BMSCs可在成骨条件培养液下诱导为成骨细胞。经鉴定,碱性磷酸酶和矿化结节染色阳性,激光共聚焦显微镜下成骨细胞鬼笔环肽显示骨架F-actin为红色荧光。结论 BMSCs在体外可定向诱导为成骨细胞,有希望成为理想的种子细胞应用于临床。
Objective To investigate the morphological and functional characteristics of osteogenic induced rabbit bone marrow mesenchymal stem cells (BMSCs) in vitro. Methods Rabbit BMSCs with good growth status were obtained in vitro and osteogenic induction was performed. The biological characteristics of osteoblasts were observed and detected. Results BMSCs could be induced into osteoblasts under osteogenic conditions. It was identified that alkaline phosphatase and mineralized nodules were positive staining, and the osteoblast phalloidin showed a red fluorescence in the framework of F-actin under laser scanning confocal microscope. Conclusion BMSCs can be induced into osteoblasts in vitro, which is an ideal seed cell for clinical application.
