目的:探讨血管活性肽Apelin及其唯一降解酶血管紧张素转换酶2(ACE2)在妊娠期高血压疾病患者及正常妊娠妇女胎盘组织中的表达。方法:分别取26例妊娠期高血压疾病妇女(妊娠期高血压10例,轻度子痫前期6例,重度子痫前期10例)(HDCP组)和28例正常妊娠妇女的胎盘组织(对照组),采用实时荧光定量逆转录聚合酶链式反应法(FO-RT-PCR)检测两组孕妇胎盘Apelin-36和ACE2 mRNA的表达情况,并用高清晰度彩色图文报告系统进行定量分析。结果:HDCP组胎盘组织Apelin-36 mRNA的表达量显著高于对照组(P“,”Objective:To explore the expressions of vasoactive peptide Apelin and its only catabolic enzyme-angiotensin converting enzyme 2(ACE2) in placental tissues of patients with hypertensive disorder complicating pregnancy(HDCP) and normal pregnant women.Methods:The placental samples of 26 HDCP women(including 10 women with gestational hypertension,6 women with mild preeclampsia,and 10 women with severe preeclampsia)(HDCP group) and 28 normal pregnant women(control group) were collected,real-time fluorescent quantitative reverse transcription-polymerase chain reaction was used to detect the expressions of Apelin-36 and ACE2 mRNA in placental tissues in the two groups,high definition color graphic report system was used for quantitative analysis.Results:The expression level of Apelin-36 mRNA in placental tissue of HDCP group was statistically significantly higher than that of control group(P<0.01);the expression level of ACE2 mRNA in placental tissue of HDCP group was statistically significantly lower than that of control group(P<0.05),which were mainly found in severe preeclampsia group(P<0.01).Conclusion:Apelin-36 expresses in placental tissue of patients with HDCP,and the increase of expression level of Apelin-36 mRNA is correlated with severity of the disease.ACE2 expresses in placental tissue of patients with HDCP,and the expression level of ACE2 mRNA is low,and the phenomenon is mainly showed in placental tissue of patients with severe preeclampsia,which indicates that ACE2 may only play an important role in onset of severe preeclampsia.There is a negative correlation between expression level of Apelin-36 mRNA and expression level of ACE2 mRNA,which indicates that degradation of Apelin-36 decreases with decrease of ACE2,and it promote the occurrence of HDCP.
