为观察姜黄醇提取物对血管平滑肌细胞增殖的影响,在用各种不同浓度姜黄酸提取物(含50%总姜黄素)处理体外培养的兔主动脉平滑肌细胞后,分别用显微镜下细胞计数法和MTT法测定细胞浓度,用丫啶橙结合台盼蓝染色观察细胞凋亡情况。结果发现136μmol.L-1姜黄素处理组的平滑肌细胞死亡率达55%,调亡细胞百分率为3%;68μmol.L-1姜黄素具有直接抑制兔平滑肌细胞增殖作用,死亡率达9.33%,凋亡细胞百分率为2.5%;当浓度为34μmol.L-1时,姜黄醇提取物没有直接抑制平滑肌细胞增殖作用,死亡率5%,凋亡细胞百分数为3.3%。但用含34μmol.L-1姜黄素及300gLDL蛋白.L-1轻度氧化LDL的培养液培养中滑肌细胞时,发现该浓度姜黄醇提取物具有抑制轻度氧化LDL促平滑肌细胞增殖的作用。
In order to observe the effect of curcumin extract on the proliferation of vascular smooth muscle cells, rabbit aortic smooth muscle cells cultured in vitro were treated with various concentrations of turmeric extract (containing 50% total curcumin), and then counted under a microscope. The cell concentration was measured by MTT assay and apoptosis was observed by acridine orange in combination with trypan blue staining. The result was found to be 136 μmol. L-1 curcumin treatment group of smooth muscle cell death rate of 55%, apoptotic cell percentage was 3%; 68μmol. L-1 curcumin had a direct inhibitory effect on the proliferation of rabbit smooth muscle cells, the mortality rate was 9.33%, the percentage of apoptotic cells was 2.5%; when the concentration was 34μmol. At the time of L-1, curcumin extract did not directly inhibit the proliferation of smooth muscle cells, the mortality rate was 5%, and the percentage of apoptotic cells was 3.3%. But with 34μmol. L-1 curcumin and 300g LDL protein. When L-1 mildly oxidized LDL was cultured in the cultured smooth muscle cells, it was found that the curcumin extract had the effect of inhibiting mild oxidized LDL to promote the proliferation of smooth muscle cells.
