目的:以H_2O_2干预大鼠肾上腺髓质嗜铬细胞瘤细胞(PC12细胞)作为模型,评价单药楮实子水煎剂对PC12细胞凋亡的影响。方法:采用MTT法检测楮实子水煎剂对PC12细胞活性的影响,比较空白组、模型组和低、中、高浓度楮实子水煎剂在H_2O_2干预下的PC12细胞生存率。应用Western Blot法检测各组Bax、Bcl-2、细胞色素C和caspase-3凋亡相关蛋白。结果:楮实子水煎剂对PC12细胞保护作用呈浓度梯度依赖增长(P<0.05)。楮实子水煎剂能阻止H_2O_2诱导PC12细胞的凋亡,并下调cleaved-caspase-3、Bax和细胞色素C蛋白的表达,呈浓度梯度依赖(P<0.05)。结论:楮实子水煎剂对H_2O_2介导大鼠肾上腺髓质嗜铬细胞瘤细胞PC12抗凋亡的作用。
OBJECTIVE: To evaluate the effects of H_2O_2 on PC12 cells in adrenal medulla in rats and to evaluate the effect of Pseudostellariae Barbata decoction on the apoptosis of PC12 cells. Methods: MTT assay was used to determine the effect of Fructus Trichosanthes decoction on the activity of PC12 cells. The survival rate of PC12 cells in blank control group, model group and low, medium and high concentration of Fructus Nisii decoction under H 2 O 2 intervention were compared. Western Blot method was used to detect Bax, Bcl-2, cytochrome C and caspase-3 apoptosis-related proteins in each group. Results: The protective effect of Fructus Lycii decoction on PC12 cells was concentration-dependently increased (P <0.05). The water decoction could prevent H 2 O 2 -induced the apoptosis of PC12 cells and down-regulated the expressions of cleaved-caspase-3, Bax and cytochrome C protein in a concentration-dependent manner (P <0.05). Conclusion: The effect of Cunjianzi decoction on the anti-apoptosis of PC12 cells induced by H 2 O 2 in rat adrenal medullary pheochromocytoma cells.
