目的体外培养未成熟畸胎瘤细胞(P 19细胞)并诱导分化,检测生殖细胞核因子(GCNF)及Oct-4在P 19细胞诱导分化前后的表达。方法用全反式维甲酸(ATRA)诱导P 19细胞分化,通过间接免疫荧光染色和RT-PCR方法检测GCNF及Oct-4在P 19细胞诱导分化前后的表达。结果 Oct-4在P 19细胞诱导前呈强阳性表达,诱导2 d后表达下降;而GCNF在P 19细胞诱导前呈阴性表达,诱导2 d后呈阳性表达。结论 GCNF参与了恶性生殖细胞肿瘤的体外分化。GCNF可能是通过下调Oct-4基因的表达参与了恶性生殖细胞肿瘤的分化。
Objective To culture and differentiate immature teratocarcinoma cells (P 19) in vitro and to detect the expression of nuclear factor of GCN (GCNF) and Oct-4 before and after induced differentiation of P 19 cells. Methods The differentiation of P 19 cells was induced by all-trans retinoic acid (ATRA), and the expression of GCNF and Oct-4 were detected by indirect immunofluorescence staining and RT-PCR before and after differentiation of P 19 cells. Results The expression of Oct-4 was strongly positive before P 19 cells induction and decreased 2 days after induction. GCNF was negatively expressed before P 19 cells induction and was positive after 2 days induction. Conclusion GCNF is involved in the in vitro differentiation of malignant germ cell tumors. GCNF may be involved in the differentiation of malignant germ cell tumors by down-regulating the expression of Oct-4 gene.
