目的探讨血必净注射液对百草枯中毒大鼠多脏器损伤的保护作用及其机制。方法Wistar大鼠54只,随机分为对照组(n=6)、染毒组(n=24)、治疗组(n=24)。染毒组和治疗组一次性胃内灌入20%百草枯溶液(50mg/kg),对照组胃内灌入等体积的生理盐水后处死留取标本。染毒后染毒组给予生理盐水4ml/kg腹腔注射,治疗组给予血必净注射液4ml/kg腹腔注射,均每天1次,连续给药7d或至处死。于第1、3、7、14天各取6只大鼠处死,留取血液及肺组织标本。采用硫代巴比妥酸比色法测定大鼠血浆丙二醛(MDA)含量,黄嘌呤氧化酶法测定超氧化物歧化酶(SOD)活力,ELISA法测定血清TNF-α及IL-10含量,肌酸激酶同工酶(CK-MB)、谷丙转氨酶(ALT)、肌酐(Cr)及淀粉酶(AMS)测定由全自动生化分析仪完成。肺组织行HE染色,光镜下观察其病理学变化。结果染毒后染毒组及治疗组MDA均明显升高,SOD明显下降,但治疗组MDA值低于染毒组,SOD值高于染毒组;2组染毒后TNF-α及IL-10均有明显升高,染毒组升高更明显;染毒后染毒组及治疗组CK-MB、ALT、Cr及AMS均有不同程度改变,但治疗组各项指标优于染毒组。光镜下染毒组于染毒后第14天可见肺组织慢性炎症改变,局部区域见纤维组织增生,间质增厚;治疗组炎症改变较轻,未见纤维化征象。结论血必净注射液可通过抗氧化及调控炎症介质作用,减轻多脏器损伤,对百草枯中毒有一定治疗作用。
Objective To investigate the protective effect of Xuebijing injection on multiple organ injury in paraquat poisoning rats and its mechanism. Methods Fifty - four Wistar rats were randomly divided into control group (n = 6), exposure group (n = 24) and treatment group (n = 24). The rats in the exposure group and the treatment group were infused with 20% paraquat solution (50 mg / kg) intragastrically. The control group was infused with an equal volume of normal saline into the stomach and sacrificed. The rats in the treated group were given intraperitoneal injection of normal saline 4ml / kg intraperitoneally, and the treatment group was injected with 4ml / kg of Xuebijing injection intraperitoneally once daily for 7 days or sacrificed. On the 1st, 3rd, 7th and 14th days, 6 rats were sacrificed and the blood and lung tissue samples were collected. Thiobarbituric acid colorimetric method was used to measure the content of malondialdehyde (MDA) in rat plasma. The activity of superoxide dismutase (SOD) was measured by xanthine oxidase method. The levels of serum TNF-α and IL-10 , Creatine kinase (CK-MB), alanine aminotransferase (ALT), creatinine (Cr) and amylase (AMS) were measured by automatic biochemical analyzer. The lung tissue was stained with HE and the pathological changes were observed under light microscope. Results After treatment, the content of MDA and the content of MDA in the treated group and the treated group were significantly decreased, but the MDA value in the treated group was lower than that in the untreated group, and the SOD value was higher than that in the untreated group. The TNF-α and IL- 10 and 10 mg · kg-1 · h-1 · h-1 · h-1 · h-1 · h-1 · h-1, respectively. . Under light microscope, the chronic inflammatory changes of lung tissue were observed on the 14th day after exposure. In the local area, fibrous tissue hyperplasia and interstitial thickening were observed. The inflammation in the treatment group changed little and there was no fibrosis sign. Conclusion Xuebijing injection can reduce the damage of multiple organs through anti-oxidation and regulation of inflammatory mediators, and has a certain therapeutic effect on paraquat poisoning.
