目的:探讨蒙古黄芪凝集素(AMML)诱导HL-60细胞凋亡的作用和机制。方法:MTT法检测AMML对HL-60细胞的增殖抑制作用。显微镜下检测细胞凋亡形态的变化,应用流式细胞仪Annexin-V/PI双染法检测细胞凋亡,同时分析细胞周期。免疫细胞化学法检测Bcl-2、Caspase-3的表达。结果:MTT法显示AMML对HL-60细胞的生长具有明显的抑制作用。荧光显微镜下,细胞呈凋亡形态学变化,流式细胞仪检测结果为细胞凋亡率明显增高,出现G0/G1期阻滞。Bcl-2表达降低,Caspase-3表达增高。结论:AMML明显抑制HL-60细胞的生长,诱导细胞凋亡。可应用于抗肿瘤药物的开发。
Objective: To investigate the effect and mechanism of Mongolia astragalus agglutinin (AMML) on apoptosis of HL-60 cells. Methods: The inhibitory effect of AMML on the proliferation of HL-60 cells was detected by MTT assay. The morphological changes of the cells were detected under the microscope. The apoptosis was detected by flow cytometry Annexin-V / PI double staining and the cell cycle was analyzed. Immunocytochemistry was used to detect the expression of Bcl-2 and Caspase-3. Results: MTT assay showed that AMML significantly inhibited the growth of HL-60 cells. Fluorescence microscopy showed morphological changes of apoptotic cells. The results of flow cytometry showed that apoptosis rate was significantly increased and G0 / G1 arrest occurred. Bcl-2 expression decreased, Caspase-3 expression increased. Conclusion: AMML significantly inhibits the growth of HL-60 cells and induces apoptosis. Can be applied to the development of anti-cancer drugs.
