建立从人外周血诱导扩增树突状细胞(DC)及吞噬凋亡小体负载抗原。方法:从正常人外周血分离获得单核细胞,加入50ng/ml rhGM-CSF 1000ng/ml rhIL—4,隔天一次,共4次,培养第三天,加入r射线照射过的胆管癌细胞,再继续体外培养一周后,用树突细胞富集柱收集DC。结果:见DC高表达共刺激分子B7和CD1a,表面具有典型不规则突起,DC还可捕捉凋亡小体、吞噬凋亡小体。负载抗原的 DC其激发同种异体 T淋巴细胞增殖的能力进一步增强。结论:用 rhGM— CSF加 rhIL— 4能从人外周血诱导、扩增 DC,此 DC可以有效提是凋亡胆管癌细胞的抗原,可望成为有效的肿瘤抗原装载DC的新途径。
The establishment of dendritic cells (DCs) induced from human peripheral blood and phagocytosis of apoptotic bodies loaded with antigens. Methods: Monocytes were isolated from normal human peripheral blood and rhIL-4 (50ng / ml rhGM-CSF, 1000ng / ml) was added once every other day for 4 times. On the third day of culture, r-irradiated cholangiocarcinoma cells were added, After a further week of in vitro culture, DCs were collected using a dendritic cell enrichment column. RESULTS: DCs co-stimulated with high expression of co-stimulatory molecules B7 and CD1a, the surface with typical irregular protrusions, DC can also capture apoptotic bodies, engulf apoptotic bodies. Antigen-loaded DC further enhances its ability to stimulate allogeneic T lymphocyte proliferation. Conclusion: rhGM-CSF plus rhIL-4 can induce and amplify DC from human peripheral blood. This DC can be an effective antigen of apoptotic cholangiocarcinoma cells and is expected to become a new effective way to load DCs with tumor antigens.
