目的研究大黄利胆胶囊对大鼠酒精性脂肪肝的影响。方法采用生理方式喂食酒精建立AFL大鼠模型,计算各组大鼠肝指数,进行肝功检测及肝组织病理学检查;生化分析法检测肝组织TG与MDA含量、抗氧化酶及ADH活性;ELISA检测血浆TNF-α含量,免疫组织化学检测肝脏CYP2E1蛋白表达。结果与模型组比较,大黄利胆胶囊低、中、高剂量组大鼠的肝指数、血清AST与ALT水平均明显降低(P<0.01或P<0.05);肝组织病理学改变减轻;TG与MDA含量均明显降低(P<0.05或P<0.01);GSH-PX均明显增加(P<0.01),SOD活性轻度增加(P>0.05);CYP2E1蛋白表达均明显下调(P<0.01);而大黄利胆胶囊中、高剂量组ADH活性明显减低及高剂量组TNF-α减少(P<0.01)。结论大黄利胆胶囊对AFL有显著的治疗作用,其作用机理可能与减轻肝脏脂肪堆积、加速乙醇清除及提高肝脏抗氧化与抗炎能力相关。
Objective To study the effect of rhubarb and li Dan capsule on alcoholic fatty liver in rats. Methods The model of AFL rats was established by physiologically feeding alcohol. The liver index of each group was calculated and the liver function and hepatic pathological examination were performed. The content of TG and MDA, the activity of antioxidant enzymes and ADH in liver tissues were detected by biochemical analysis. The level of plasma TNF-α was detected and the expression of CYP2E1 protein in liver was detected by immunohistochemistry. Results Compared with the model group, the liver index, serum AST and ALT levels were significantly decreased (P <0.01 or P <0.05), and the pathological changes of liver tissue were relieved in TG, (P <0.05 or P <0.01). The levels of GSH-PX were significantly increased (P <0.01) and the activities of SOD were slightly increased (P> 0.05). The protein expression of CYP2E1 was significantly decreased (P <0.01) However, the activity of ADH in rhubarb and Lactobacillus in the high dose group was significantly decreased and the TNF-α in high dose group was decreased (P <0.01). Conclusion Rhubarb Lactobacillus capsule has a significant therapeutic effect on AFL, and its mechanism may be related to reducing hepatic fat accumulation, accelerating ethanol clearance and improving liver anti-oxidation and anti-inflammatory abilities.
