目的:探讨银花合剂对急性肺损伤(ALI)大鼠干预作用与可能机制。方法:采用油酸加内毒素“二次打击”方法复制大鼠ALI模型,用银花合剂进行干预,实验结束后测定动脉血氧分压、肺湿/干质量比值、肺病理形态学积分、血浆IL-1β的含量及大鼠海马CA2-3区FOS蛋白阳性细胞灰度值。结果:与对照组比较,模型组大鼠IL-1β含量显著升高(P<0.01),海马FOS阳性细胞灰度值迅速下降(P<0.01),与模型组相比,银花合剂组和地塞米松组氧分压上升,肺湿/干质量比值、病理形态学积分、IL-1β含量显著下降(P<0.01),FOS蛋白阳性细胞灰度值明显增高(P<0.01)。结论:银花合剂能够减轻肺损伤,其可能机制是银花合剂通过拮抗IL-1β,抑制海马c-fos表达实现对急性肺损伤大鼠的防治作用。
Objective: To investigate the effects and possible mechanisms of Yinhua mixture on acute lung injury (ALI) in rats. Methods: The rat model of ALI was reproduced by oleic acid plus endotoxin “second strike” method, and treated with Yinhua mixture. After the experiment, the arterial partial pressure of oxygen, the ratio of lung wet / dry weight, the pathological morphology of lung Integral, plasma IL-1β content and FOS protein positive cells in hippocampal CA2-3 area. Results: Compared with the control group, the content of IL-1β in the model group was significantly increased (P <0.01), and the gray value of FOS positive cells in the hippocampus decreased rapidly (P <0.01). Compared with the model group, Dexamethasone group increased oxygen partial pressure, lung wet / dry mass ratio, pathological score, IL-1β content decreased significantly (P <0.01), FOS protein positive cell gray value was significantly increased (P <0.01). CONCLUSION: Yinhua Mixture can reduce lung injury. Its possible mechanism is that Yinhua Mixture can prevent and treat acute lung injury in rats by antagonizing IL-1β and inhibiting the expression of c-fos in hippocampus.
