目的探讨Tiam1在正常乳腺组织、乳腺癌组织及不同侵袭能力乳腺癌细胞系中的表达情况。方法应用免疫组化方法检测Tiam1在60例乳腺癌组织和34例正常乳腺组织中的表达。应用Westernblot方法检测Tiam1在乳腺癌细胞系MDA-MA-231(高侵袭)和MCF-7(低侵袭)中的表达。结果Tiam1阳性率在正常乳腺组织(41%)中明显低于乳腺癌组织(71.7%,P<0.01),Tiam1在乳腺癌组织中的表达与肿瘤的TMN分期、是否发生淋巴结转移以及病理分级密切相关(P<0.05)而与肿瘤的大小无关。Tiam1总蛋白在乳腺癌高侵袭细胞系中的表达量高于低侵袭细胞系,差异具有显著性(P<0.05)。结论Tiam1在乳腺癌中的表达与其转移侵袭相关,可能是判断预后的一个潜在指标。
Objective To investigate the expression of Tiam1 in normal breast tissues, breast cancer tissues and different invasive breast cancer cell lines. Methods Immunohistochemistry was used to detect the expression of Tiam1 in 60 cases of breast cancer tissues and 34 cases of normal breast tissues. The expression of Tiam1 in breast cancer cell lines MDA-MA-231 (high invasion) and MCF-7 (low invasion) was detected by Western blot. Results The positive rate of Tiam1 was significantly lower in normal breast tissue (41%) than that in breast cancer (71.7%, P <0.01). The expression of Tiam1 in breast cancer tissues was related to TMN stage, lymph node metastasis and pathological grading (P <0.05) but not with tumor size. The expression of Tiam1 protein in breast cancer cell lines with high invasiveness was higher than that in low invasive cell lines (P <0.05). Conclusion The expression of Tiam1 in breast cancer is related to its metastasis and invasion, which may be a potential indicator of prognosis.
