目的:采用固相萃取-高效液相色谱法同时测定食品中七种合成色素(柠檬黄、苋菜红、靛蓝、胭脂红、日落黄、诱惑红、亮蓝)。方法:样品经水溶解后,选用StrataX-AW SPE弱阴离子小柱净化,HPLC测定。色谱条件:Agilent C18柱(250 mm×4.6 mm,5μm),流动相:甲醇-0.02 mol/L磷酸二氢钾水溶液(用NaOH调至pH5.8),流速1.0 ml/min,进样量10μl,柱温35℃。检测波长:428 nm,520 nm,620 nm,梯度洗脱。结果:七种色素在0.20 mg/L~5.00 mg/L范围内与响应值呈线性关系,不同水平加标回收率为85%~110%,RSD为2.1%~4.8%,最低检出浓度均小于或等于0.001g/kg。结论:方法简便、快速、灵敏、准确,适用于食品中七种色素的同时测定。
OBJECTIVE: To simultaneously determine seven kinds of synthetic pigments (lemon yellow, amaranth, indigo, carmine, sunset yellow, seductive red and bright blue) in foods by solid-phase extraction and high performance liquid chromatography. Methods: The samples were dissolved in water and purified by StrataX-AW SPE weak anion column and determined by HPLC. The chromatographic conditions were as follows: Agilent C18 column (250 mm × 4.6 mm, 5 μm), mobile phase: methanol 0.02 mol / L potassium dihydrogen phosphate (adjusted to pH 5.8 with NaOH), flow rate 1.0 ml / , Column temperature 35 ℃. Detection wavelength: 428 nm, 520 nm, 620 nm, gradient elution. Results: The linear range of the seven pigments was 0.20 mg / L ~ 5.00 mg / L with the recoveries of 85% -110% and RSD of 2.1% -4.8% at different levels. The minimum detectable concentrations Less than or equal to 0.001 g / kg. Conclusion: The method is simple, rapid, sensitive and accurate and suitable for simultaneous determination of seven pigments in food.
