目的:研究丹参通脉方对同型半胱氨酸(HCY)诱导增殖的人主动脉血管平滑肌细胞(HA-VSMC)内质网应激(ERS)相关凋亡通路的影响,探讨丹参通脉方的心血管药理学作用机制。方法:应用HCY诱导HAVSMC增殖,使用丹参通脉方干预,Annexin V-FITC/PI法检测细胞凋亡,Western blot及实时荧光定量PCR法检测ERS通路中免疫球蛋白结合蛋白(BIP)、C/EBP同源蛋白(CHOP)蛋白及基因表达。结果:与空白组及模型组比较,丹参通脉方促进HA-VSMC凋亡;丹参通脉方可促进BIP、CHOP蛋白及基因的表达(P<0.05)。结论:丹参通脉方可促进异常增殖的HA-VSMC凋亡,其作用机制可能与促进ERS相关凋亡通路信号转导有关。
Objective: To investigate the effect of Danshen Tongmai Recipe on the apoptosis pathway of endoplasmic reticulum stress (ERS) in human aortic vascular smooth muscle cells (HA-VSMCs) induced by homocysteine (HCY) Cardiovascular pharmacological mechanism of action. Methods: Proliferation of HAVSMC was induced by HCY. The apoptosis of HAVSMC was detected by using Danshen Tongmai Fang intervention and Annexin V-FITC / PI. Western blot and real-time fluorescence quantitative PCR were used to detect the expression of immunoglobulin binding protein (BIP), C / EBP homologous protein (CHOP) protein and gene expression. Results: Compared with the blank group and the model group, Danshen Tongmai Fang could promote the apoptosis of HA-VSMC; and Danshen Tongmai could promote the expression of BIP, CHOP protein and gene (P <0.05). Conclusion: Danshen Tongmai can promote apoptosis of HA-VSMCs with abnormal proliferation. Its mechanism may be related to the signal transduction of apoptosis pathway related to ERS.
