采用水提醇沉法从云南丽江产白雪茶中分离提取出雪茶多糖(Thamnolia vermicularis polysaccharide,TP),经石油醚、乙醇回流去除脂类、小分子杂质、Sevag法脱蛋白、透析后,用硫酸-苯酚法测定了多糖含量。结果表明,在10μg/ml-80μg/ml范围内,吸光度与被测物含量之间具有较好的线性关系(R2=0.9982)。重复性试验、回收率试验表明,该方法具有较好的重复性(RSD=2.47%)与准确度(RSD=2.76%)。测定结果表明,雪茶多糖中总糖含量为81.26%(RSD=2.13%),雪茶原料中多糖含量为2.78%。
Thamnolia vermicularis polysaccharide (TP) was isolated from Baiyue tea produced in Lijiang, Yunnan by water extraction and alcohol precipitation method. Lipids and small molecular impurities were removed by refluxing with petroleum ether and ethanol. The proteins were deproteinized by Sevag method. After dialysis, Sulfate - phenol method for the determination of polysaccharide content. The results showed that there was a good linear relationship between absorbance and analyte content in the range of 10μg / ml-80μg / ml (R2 = 0.9982). Repeatability tests and recovery tests show that the method has good repeatability (RSD = 2.47%) and accuracy (RSD = 2.76%). The results showed that the total content of polysaccharide in Xuecha polysaccharide was 81.26% (RSD = 2.13%), and the content of polysaccharide in Xuecha raw material was 2.78%.
