背景:神经安定类药物广泛应用于临床,但各药对气道平滑肌的松弛效应尚无比较研究。目的:为了对气道高反应性患者筛选最佳神经安定药物,对不同浓度的安定、氟哌利多、氯丙嗪及异丙嗪对离体兔气管平滑肌的松弛效应进行了比较研究。设计:自身对照实验研究。地点和对象:实验在南京医科大学第一附属医院麻醉科完成,成年健康兔24只,雌雄各12只,体质量(2.5±0.5)kg,由南京医科大学实验动物研究中心提供。干预:将兔随机分为4组。将离体兔气管平滑肌标本浸于35,140μmol/L安定,2.6,10.4μmol/L氟哌利多,28,112μmol/L氯丙嗪及31,124μmol/L异丙嗪中,用电场刺激测定离体兔气管平滑肌张力。自身用药前后比较采用t检验。主要观察指标:安定、氟哌利多、氯丙嗪及异丙嗪对电场刺激引起的兔气管平滑肌张力变化。结果:4种药物均使兔气管平滑肌张力呈浓度依赖性减弱,但低浓度安定时无显著统计学意义(t=2.568,P>0.05),高浓度安定用药前后平滑肌张力分别为(430±264)和(297±212)mg,有显著的统计学意义(t=2.821,P<0.05);高、低浓度氟哌利多、氯丙嗪及异丙嗪均有显著或非常显著统计学意义(t>2.921,P<0.05,或t>4.125,P<0.01)。等效镇静剂量,各药降低气管平滑肌收缩强度依次为异丙嗪>氯丙嗪>氟哌利多>安定。结论:气道高反应性患
BACKGROUND: Neuroleptics are widely used in clinical practice. However, the relaxation effects of various drugs on airway smooth muscle have not been compared. OBJECTIVE: To screen the best neuroleptic agents for patients with airway hyperresponsiveness, the relaxation effects of different concentrations of diazepam, droperidol, chlorpromazine and promethazine on tracheal smooth muscle of rabbits were compared. Design: self-control experimental study. Location and Subjects: The experiment was performed in the Anesthesiology Department of the First Affiliated Hospital of Nanjing Medical University. Twenty-four healthy rabbits were male and 12 females with a body weight of (2.5 ± 0.5) kg provided by Experimental Animal Research Center, Nanjing Medical University. Interventions: Rabbits were randomly divided into 4 groups. The isolated rabbit bronchial smooth muscle specimens were immersed in 35,140μmol / L diazepam, 2.6,10.4μmol / L droperidol, 28,112μmol / L chlorpromazine and 31,124μmol / L promethazine. The tension of smooth muscle before and after diazepam was (430 ± 264) ) And (297 ± 212) mg respectively (t = 2.821, P <0.05). There was significant or very significant difference between high and low concentrations of droperidol, chlorpromazine and promethazine t> 2.921, P <0.05, or t> 4.125, P <0.01).
